Leigh Syndrome

Leigh Syndrome is a disease of impaired energy metabolism where both copies of the inherited LRPPRC gene are mutated. This results in brain damage, delayed childhood development, incoordination of movement, and liver complications. Lactic acidosis can be fatal; otherwise, death occurs between six months and 12 years of age.

It has been believed that T lymphocytes of the immune system learn tolerance (avoiding attack) on self-tissues by confronting their native proteins in two areas:

  1. the thymus gland, and
  2. peripheral tissues.

However researchers, at Joslin Diabetes Center, Harvard Medical School, and other institutions, have recently shown that T cells confront the peripheral self-proteins in the thymus gland itself.

These "shadow proteins" result from the expression of numerous genes associated with the liver, brain, pancreas and other organs.
